Brief Introduction
5-hydroxymethyltolterodine is a major active metabolite of tolterodine. 5-hydroxymethyltolterodine competitively inhibited the contraction of isolated guinea pig bladder strips induced by carbachol in a concentration dependent manner. The pa2 value of 5-hydroxymethyltolterodine against muscarinic receptor was 9.1. 5-hydroxymethyltolterodine can inhibit (-) - 5-hydroxymethyltolterodine in the homogenate of bladder, parotid gland, heart and cerebral cortex of guinea pigs, which has similar pharmacological properties to tolterodine. Intravenous injection of 5-hydroxymethyltolterodine inhibited bladder contraction induced by bladder volume, which represents voiding pressure, in a dose-dependent manner.
