Brief Introduction
Glibenclamide, also known as youjiangtang and daantai, belongs to the second generation of oral sulfonylurea hypoglycemic drugs. Its action mechanism is the same as tolbutamide. Its hypoglycemic effect is the strongest among sulfonylureas, and its action intensity is 200 ~ 250 times that of tolbutamide. It can selectively act on the pancreas β Cells and promote the secretion of insulin, which can enhance the hypoglycemic effect of exogenous insulin and strengthen the post receptor effect of insulin. Oral absorption is fast and the protein binding rate is high. The effect begins when the chemical book is opened in 30 minutes, and the effect is the strongest in 1.5 hours, lasting for 16 ~ 24 hours. The distribution volume is 0.1l/kg, the plasma protein binding rate is 90% ~ 95%, and the half-life is 4 ~ 8 hours. It is mainly metabolized by the liver. There are six metabolites, two of which are known to be hydroxylates, which have no hypoglycemic effect. They are mainly discharged from the urine and a small amount from the stool. Clinically, it is mainly used to treat mild to moderate non-insulin-dependent diabetes mellitus.
